Белок CRM197 является перспективным адъювантом нового поколения, который может быть успешно использован в вакцинных и терапевтических препаратах. Классическим способом получения дифтерийного токсина и его нетоксичных вариантов является продукция в клетках Corynebacterium diphtheria. Преимущество использования E.coli в качестве продуцента заключается в том, что данный метод является более простым и дешевым, и позволяет получать рекомбинантный CRM197 в короткие сроки с использованием непатогенного микроорганизма. В ходе исследования разработан состав кандидатной субстанции CRM197 на основе анализа физико-химических свойств белка, а также специфики его применения в составе конъюгированных вакцин. В субстанции рекомбинантного белка CRM197 присутствуют примеси белка штамма-продуцента - 0,054 нг/мкг белка CRM197, ДНК штамма-продуцента 1,26 пг/мкг белка CRM197, а также остаточные эндотоксины менее 0,078 EU на 1 мг белка. Таким образом, белок CRM197, являющийся нетоксичным вариантом дифтерийного токсина, представляет собой перспективный адъювант нового поколения, который может быть успешно использован в вакцинных и терапевтических препаратах. Использование трехступенчатой очистки позволило получить рекомбинантный белок CRM197 фармацевтически приемлемого качества. Данный метод имеет перспективы по внедрению в производственную практику.</p></abstract><trans-abstract xml:lang="en"><p>CRM197 is a non-toxic mutant of diphtheria toxin having a single amino acid substitution of a glycine for a glutamic acid in position 52. Being naturally nontoxic, CRM197 is a promising adjuvant and ideal carrier protein for conjugate vaccines. Typically, production of diphtheria toxic and non-toxic mutants are carried out in Corynebacterium diphtheria. Production of recombinant CRM197 in Escherichia coli is advantageous. It is simple, cheap and permits production of the target protein in a short time using a non-pathogenic microorganism. In this study the pharmacology grade substance of recombinant protein CRM197 was developed based on chemical properties of the protein.
