Эпидемиологическая обстановка в мире характеризуется увеличением заболеваемости людей и животных ортопоксвирусными инфекциями. В связи с этим необходимо разрабатывать новые безопасные вакцины против этих инфекций.</p></sec><sec><title>Цель</title><p>Цель. Проведение доклинических исследований специфической активности вакцины против оспы и других ортопоксвирусных инфекций VACΔ6 на основе вируса осповакцины вируса осповакцины (ВОВ) с шестью нарушенными генами вирулентности.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Исследования выполнены в соответствии с требованиями Руководства по проведению доклинических исследований лекарственных средств (иммунобиологические препараты), Государственной фармакопеи ХIII и Европейской Фармакопеи 7.0.</p></sec><sec><title>Результаты и обсуждение</title><p>Результаты и обсуждение. Вакцинный штамм VACΔ6 ВОВ на модели внутримозгового введения мышам­сосункам проявил значительно уменьшенную нейровирулентность, а на модели внутрикожного введения кроликам – сниженную воспалительно­-некротическую активность по сравнению с классической оспенной живой вакциной первого поколения, разрешённой для использования в России. Доклинические исследования трёх серий готовой лекарственной формы вакцины VAC∆6 показали её подлинность, термостабильность, апирогенность и безвредность. При двукратной внутрикожной вакцинации кроликов в дозе 106 оспообразующих единиц (ООЕ) обеспечивался 100% защитный эффект против интраназального инфицирования кроликов ВОВ штамм НВ­92 в дозе 4,9 lg бляшкообразующих единиц (БОЕ) (1995 ЛД50) и при двукратной внутрикожной вакцинации мышей в дозе 106 ООЕ обеспечивалась полная защита против интраназального инфицирования мышей вирусом эктромелии штамм K1 в дозе 3,05 lg БОЕ (56 ЛД50).</p></sec><sec><title>Вывод</title><p>Вывод. На основании проведённого комплекса исследований можно заключить, что созданная вакцина четвёртого поколения VACΔ6 является более безопасной по сравнению с Вакциной оспенной живой первого поколения и не уступает ей по иммуногенным и протективным свойствам.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Relevance</title><p>Relevance. The epidemiological situation in the world is characterized by an increase in the incidence of orthopoxvirus infections in humans and animals. In this regard, it is necessary to develop new safe vaccines against these infections.</p></sec><sec><title>Aim</title><p>Aim. Conducting preclinical studies on the specific activity of the live vaccine against smallpox and other orthopoxvirus infections VACΔ6 based on the vaccinia virus (VACV) with six deleted virulence genes are presented.</p></sec><sec><title>Matherials and methods</title><p>Matherials and methods. The studies were performed in accordance with the requirements of the Guidelines for conducting preclinical studies of drugs (immunobiological preparations), the State Pharmacopoeia XIII and the European Pharmacopoeia 7.0.</p></sec><sec><title>Results and discussion</title><p>Results and discussion. The vaccine strain VACΔ6 VACV showed significantly reduced neurovirulence in the model of intracerebral administration to suckling mice, and reduced inflammatorynecrotic activity in the model of intradermal administration to rabbits compared to the classical first­generation smallpox live vaccine approved for use in Russia. Preclinical studies of three series of the finished dosage form of the VAC∆6 vaccine showed its authenticity, thermal stability, non­pyrogenicity and safety. Double intradermal vaccination of rabbits at a dose of 106 PFU/animal, a 100% protective effect was provided against the intranasal infection of rabbits with VACV strain HB­-92 at a dose of 1995 LD50 and a double intradermal vaccination of mice at a dose of 106 PFU/animal, full protection was provided against the intranasal infection of mice with ectromelia virus strain K1 at a dose of 56 LD50.</p></sec><sec><title>Conclusion</title><p>Conclusion.
