Listeria monocytogenes – повсеместно распространенная бактерия, возбудитель листериоза. Это широко распространенная инфекционная болезнь, которая в настоящее время наносит большой ущерб животноводству и является серьезной угрозой здоровью людей.</p></sec><sec><title>Цель</title><p>Цель. Проанализировать популяционную структуру и оценить патогенный потенциал изолятов Listeria monocytogenes, выделенных на территории РФ.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Всего из продуктов питания было выделено 79 изолятов листерий. Их видовую идентификацию и фенотипический анализ на устойчивость к антибиотикам проводили на приборе VITEK MS system (bioMerieux, Marcyl’toile, Франция). Тридцать пять антибиотикорезистентных изолятов были охарактеризованы с помощью анализа данных полногеномного секвенирования.  ь антибиотикорезистентных изолятов были охарактеризованы с помощью анализа данных полногеномного секвенирования. Результаты. Определена клональная структура изученной популяции, выявлены немногочисленные детерминанты устойчивости к антибиотикам (fosX, tetM и сlpL), обширный набор факторов вирулентности и системы CRISPR/Cas. Большая часть изолятов относилась ко II филогенетической линии и разделилась на девять клональных комплексов с доминированием СС 121, который является одним из эпидемиологически значимых генетических клоном. Обнаружены также два СС2-изолята, принадлежащих к наиболее патогенной филогенетической линии I. Тринадцать изолятов характеризовались наличием полноценных CRISPR/Cas систем IB и IIA типов, при этом все СС121-изоляты в своих геномах содержали одновременно два типа выявленных систем адаптивного иммунитета. Корреляционный анализ подтвердил их функциональность.</p></sec><sec><title>Заключение</title><p>Заключение. Мы полагаем, что полученные данные о полных геномах изолятов Listeria monocytogenes пищевого происхождения облегчат и дополнят дальнейшие исследования эпидемиологии данного патогена, вариабельности его генома с точки зрения приобретения различных генетических элементов, связанных с адаптацией, устойчивостью к противомикробным препаратам и вирулентностью. В свою очередь, результаты таких исследований помогут разработать профилактические меры для эффективного решения проблем, связанных с контаминацией бактериальными патогенами продуктов животного происхождения, и обеспечения пищевой безопасности в условиях производств и векторной цепочки «от фермы до стола».</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Relevance</title><p>Relevance. Listeria monocytogenes is a ubiquitous bacterium that causes listeriosis, which represents a widespread infectious disease currently inflicting great damage to livestock production and posing a serious threat to human health.</p></sec><sec><title>Aim</title><p>Aim. To analyze the population structure and assess the pathogenic potential of Listeria monocytogenes isolates isolated on the territory of the Russian Federation.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. A total of 79 listeria isolates were isolated from food products. Species identification and phenotypic analysis for antibiotic resistance were performed using VITEK MS system (bioMerieux, Marcyl’toile, France). Thirty-five antibiotic-resistant isolates were characterized by analysis of whole-genome sequencing data.</p></sec><sec><title>Results</title><p>Results. Whole genome sequences of thirty-five antibiotic-resistant Listeria monocytogenes isolates of food origin were analyzed. We determined clonal structure of this population and revealed a small number of antibiotic resistance determinants (fosX, tetM и сlpL), extensive set of virulence factors, as well as the presence of CRISPR/Cas systems. Most of the isolates belonged to phylogenetic line II and were divided into nine clonal complexes with the prevalence of CC121, which was one of the epidemiologically significant genetic clones. Two CC2 isolates belonging to the most pathogenic phylogenetic lineage I were also found. Thirteen isolates were characterized by the presence of putative CRISPR/Cas systems of IB and IIA types. All ST 121 isolates contained two types of identified adaptive immunity systems simultaneously in their genomes. Correlation analysis confirmed their functionality.</p></sec><sec><title>Conclusion</title><p>Conclusion. We believe that the whole genome data obtained for the foodborne Listeria monocytogenes isolates will facilitate and complement further epidemiological studies of this pathogen, as well as the investigations of its genome variability in terms of the acquisition of various genetic elements associated with adaptation, antimicrobial resistance, and virulence.
